What is Melanotan 2?
Melanotan 2 (often abbreviated as MT-2 or Melanotan II) is a synthetic peptide hormone designed to mimic the effects of alpha-melanocyte-stimulating hormone (α-MSH), a natural substance in the body that regulates skin pigmentation. Originally developed in the 1980s by researchers at the University of Arizona, it was intended to stimulate melanin production for tanning without excessive UV exposure, potentially reducing skin cancer risk. However, development for cosmetic use was largely abandoned in the 2000s due to regulatory hurdles and safety concerns.
Unlike Melanotan 1 (afamelanotide, an FDA-approved drug for specific light-sensitive skin conditions like erythropoietic protoporphyria), Melanotan 2 is not approved for any medical use by major regulatory bodies such as the FDA, TGA (Australia), or HPRA (Ireland). It’s widely available online as an unregulated injectable, nasal spray, or powder, often marketed for “tanning without the sun,” but this is illegal in many countries and carries significant health risks.
How Does It Work?
Melanotan 2 acts as a non-selective agonist for melanocortin receptors (especially MC1R and MC4R) in the skin, brain, and other tissues. This leads to:
- Increased melanin production: Darkens the skin for a tanned appearance, even in fair-skinned individuals, with effects noticeable after minimal sun exposure.
- Other effects: It can suppress appetite, boost libido, and induce erections by influencing central nervous system pathways.
The peptide is typically reconstituted with bacteriostatic water and administered via subcutaneous injection (e.g., 0.25–1 mg doses) or nasal spray. Synthesis involves a 12-step process yielding about 2.6% overall, resulting in a product that’s over 90% pure without advanced purification.
Potential Benefits
While unproven for general use and not recommended, research and user reports suggest:
- Tanning: Rapid skin darkening (within days) with less UV need, potentially lowering sunburn risk.
- Appetite suppression: May aid weight loss by reducing hunger via MC4R activation.
- Sexual enhancement: Increases libido and erectile function in both men and women; a derivative, bremelanotide (Vyleesi), is FDA-approved for hypoactive sexual desire disorder in women.
- Other research areas: Explored for erectile dysfunction, but largely replaced by safer alternatives.
